Introduction

The present study examined the acute myocardial ischemia with Chinese herbs complex prescription, Cleaning Lipid Tabellae (CLT), researched the effect and mechanism of on anti-myocardial ischemia.

Methods

Wistar rats were divided into 4 groups including control, model, low dose CLT and high dose groups with 10 rats each. Rats from both genders were equally divided into the groups. CLT intervention was carried by infusing into the stomach once every day, 7 days before making acute myocardial ischemic by injecting pituitrinum into the tail vein[1] twice. Researching indices consisted of lead II ECG, serum creatine phosphokinase (CPK), the platelet adhesion rate, enlarged platelet rate, the number of aggregation, myocardium NOS mRNA and HSP70 mRNA gene expression with the techniques of in site hybridization and image pattern analysis.

Results

Lead II ECG recordings showed that, in 15 minutes after the second injection of pituitrinum, ST-T ischemic changes were observed in the model group with significant increase in level of serum creatine phosphokinase (CPK). CLT significantly reduced the ST-T ischemic changes of ECG, ischemic positive rate and the activity of serum CPK in both high dose and low dose compared with the model group. The platelet adhesion rate, enlarged platelet rate and the number of aggregation were decreased in both groups compared with the model group. The results revealed that myocardial ischemia could induce the NOS mRNA expression. However, high dose CLT could cause over expression of myocardial cell NOS mRNA.

Discussion/Conclusion

The results supported the effects of CLT in preventing and treating myocardial ischemia, protecting myocardial cytomembrane, reducing the CPK leakage from myocardial cell, restraining platelet adhesion and aggregation, and maintaining platelet normal form and function [2]. One of the potential mechanisms to resist myocardial ischemia is to switch on myocardial cell self-protecting mechanism by increasing NO[3]. Myocardial ischemia also induced HSP70 mRNA expression. Over expression of HSP70 mRNA with high dose CLT was observed. CLT might maintain normal structure and homeostasis of myocardial cell and prevent myocardial ischemia[4].
